Mitotane, or o,p'-DDD, is a derivative of DDT (sc-238975) with antiproliferative properties. A study of the antiproliferative effects of
Mitotane demonstrated particular efficacy against adrenocortical carcinoma.
For the treatment of adrenocortical carcinoma and its migratory cancer, it can also be used for adrenal hyperplasia or tumor-induced hypercortisolism
Mitotanthan white crystalline solid. The molecular formula is C14H10Cl4, and the molecular weight is 320.04100. The melting point is 76-78 °C. Soluble in ethanol and carbon tetrachloride. The density is 1.372 g/cm3, the melting point is 77-78 °C (lit.), the boiling point is 398.9oC at 760 mmHg, the flash point is 194.2oC, and the warehouse is ventilated and dried at low temperature.
1. 6~15mg/ per day, divided into 3~4 times orally, gradually increase from small dose to maximum tolerance, the range of change is 2~16g per day, generally 8~10g per day. 2. If an adverse reaction occurs, reduce the dose until the maximum tolerated dose is determined. The treatment should continue until the clinical effect occurs. After the treatment, it is changed to 2~4g per day, and 4~8 weeks is a course of treatment. If the maximum tolerated dose is still invalid for 3 months, the treatment is stopped. Treatment should be performed under the direction of a physician familiar with the drug.
